Recent discoveries have led to new insights into the biology of this virus that may aid in the development of an effective vaccine or drug. For example, the virus encodes a fusion glycoprotein, F, and a second viral glycoprotein, G, that are crucial for the fusion of the virus with the host cell membrane. The F protein, which is structurally and functionally conserved among RSV strains, is the major target for neutralizing antibody. However, the antibody response to the F protein elicited by natural infection is mainly strain-specific. It appears that the F protein can induce broadly neutralizing antibody responses to different strains of RSV. In order to elucidate the mechanism by which the F protein induces broadly neutralizing antibody, and to identify other targets on the F protein for inducing broadly neutralizing antibody, we have developed a unique animal model for studying the F protein. This model, called BALB/c-ma, allows the rapid evaluation of F protein immunogens and analysis of the immune response to the F protein. The unique properties of this animal model will be used to address questions about the F protein, including: 1) Which immunodominant regions on the F protein induce broadly neutralizing antibodies; 2) How does the F protein induce broadly neutralizing antibody?; and 3) How do neutralizing antibodies block infection by the virus?
